The DNA of the phage phi80ilambdadhis, which carries the histidine operon (his) of E. coli K12, has been used as template for transcription in vitro catalyzed by E.coli RNA polymerase. The product obtained has been charcterized through hybridization to the DNA of the phages phi80ilambdadhis and phi80ilambda. Hybridization to the separated strands and hybridization competion show that the his operon is actually transcribed in vitro on the correct DNA strand.
